You shouldn't have to remove too much stuff to get to them. If your car is the same set up as the 65-67 series then all you'll have to take out is the back seat bench, handles, and quarter trim panel. You may need to loosen the door jamb scuff molding to get the qarter trim panel out too.
Also, you may need to remove (or just loosen) the rear-most section of the window run channel to get the old regulator out and the new one in.
All in all, no big deal. Don't forget to grease 'em up before you put them in, it's hard to do it afterwards.
