Yes you have to plug the tail end of the tranny or fluid will spill out. I always just used a plastic bag and a zip tie to cover it. You could leave the tranny in but if it's your first time and you're doing it in the driveway it'll be easier to get it out of the way so you have more room to work. I always take the tranny out, but I know guys that work at dealerships that don't, to save time, when changing clutches and rear mains etc.
